Other growth factor/hormone receptors that signal through the IRS proteins and that are associated with cancer include the insulin, prolactin, growth hormone (GH), and vascular endothelial growth factor (VEGF; KDR) receptors .

Accordingly, we found that all but one of the 17 BRCA1tumors contained CD44+/CD24- cells, further illustrating the correlation between CD44+ status and basal-like/BRCA1-like tumors.
Immunohistochemical features have been used to characterize basal-like tumors as typically negative for ER, for the progesterone receptor (PgR) and for HER2 but positive for basal cytokeratins (CK5/6/14/17), for epidermal growth factor receptor (EGFR) and/or for c-kit .
The luminal subtypes are associated with expression of the estrogen receptor (ER), while basal-like and normal-like tumors are essentially all ER-negative, as are the majority of HER2+ tumors.
Furthermore, APRIL that is produced in tumors is retained and accumulates in the tumor stroma though HSPG binding.

BAFF is overexpressed in Hodgkin lymhoma compared to normal B cells (P = 4.9E-11), in Burkitt lymphoma compared to normal B cells (P = 5.4E-5), in diffuse large B cell lymphoma compared to normal B cells (P = .01) and in follicular lymphoma compared to normal B cells (P = 2E-6).
APRIL was overexpressed in invasive bladder carcinoma compared to superficial bladder carcinoma in two independent studies (P = 3.3E-6 and P = 0.003), in esophageal adenocarcinoma compared to normal esophagus in two independent studies (P = 3.7E-4 and P = 5.5E-4), in glioblastoma compared to normal brain in two independent studies (P = 3.5E-4 and P = 0.006), in head and neck carcinoma compared to normal oral mucosa in two independent studies (P = 9.2E-10 and P = 0.009), in diffuse large B cell lymphoma compared to normal lymph nodes (P = 7.6E-6), in pancreatic adenocarcinoma compared to normal pancreas (Iocobuzio-Donahue et al; P = 0.002) and in adult male germ cell tumor compared to normal testis (Korkola et al; P = 1.6E-11) (Figure ).
Fig shows that the density was indeed significantly higher in VEGF-C overexpressing tumors compared with PC-3/mock tumors (220 ± 15 μm/mm2 vs. 37 ± 6 μm/mm2, p < 0.001).
As in the subcutaneous tumors, the orthotopic tumors overexpressing VEGF-C were markedly bigger (400 ± 17 mm3 vs. 48 ± 4 mm3, p < 0.001, Fig ).
Next, we studied if we can block the VEGFR3 pathway in VEGF-C overexpressing orthotopic tumors.
Regulation of VEGF-C and VEGFR3 in prostate cancer metastasis was studied using orthotopic PC-3prostate tumors, which naturally express VEGF-C.
These data suggest that high cytokine levels are associated with poor prognostic factors such as high grade, ER-negative status, and HER2-positive tumours.
The cytokines IL-5, IL-17 and GM-CSF were not detected in either normal or tumour sample, whereas IL-1β, IL-2, IL-4, G-CSF and IL-10 were only detected in breast carcinoma.
Among these cytokines, the greatest differences were observed for IL-8 and MCP-1, which respectively exhibited 3.9-fold and 2.7-fold higher expression in AP-1-positive tumours than in AP-1-negative ones (Table ).
On the other hand, IL-2, IL-6, IL-8, IL-10, IFN-γ, MCP-1, MIP-1β, TNF-α and, to a lesser extent, IL-1β and IL-13 were significantly overexpressed in ER-negative tumours compared with ER-positive ones, with the greatest differences observed for IL-8 and MCP-1.
Consistent with these observations and recent reports , HER2+ tumors in the current study showed intermediate expression of two ER gene expression modules, which were significantly less expressed in HER2- basal-like tumors.
The ability of the BAC aCGH platform to accurately estimate HER2 copy numbers was confirmed by parallel FISH analysis (Additional file ) in 13 FFPE HER2-amplified tumors, showing a good correlation between the techniques (Figure S1A in Additional file ).
In 31% of HER2+ tumors the amplicon extended to TOP2A, defining a subgroup of HER2+ breast cancer associated with estrogen receptor-positive status and with a trend of better survival than HER2+ breast cancers with deleted (18%) or neutral TOP2A (51%).
HER2-amplified (HER2+) tumors define a clinically important BC subgroup, generally associated with poor prognosis .
